Abstract

Verbascoside, also known as Acteoside, has been found in over 23 families of plants. demonstrating, among many other properties, utility in vivo as an antihypertensive. However, its effect on the vascular system is not fully known. This work aims to study the vascular reactivity of verbascoside on rat aorta, hypothesizing a possible vascular relaxing effect responsible for the antihypertensive activity. The concentration-response curves were carried out on 3mm long rings of rat aorta, evaluating the response to verbascoside and phenylephrine, acetylcholine, sodium nitroprusside and calcium in the presence of verbascoside at different molar concentrations (10-6, 10-5 y 10-4 M). The results obtained show that verbascoside, in vitro, does not have a vascular relaxing effect and does not affect the basal tone of the aorta, it does not enhance the contracting action of phenylephrine, but nevertheless it does reduce the vascular relaxation derived from acetylcholine. On the other hand, it slightly enhances the vascular relaxing action of nitroprusside, not interfering with the entry of extracellular calcium. Thus, the antihypertensive in vivo activity of verbascoside is not explained by the in vitro activity seen in these experiments.
